Background of Synaptopodin

Synaptopodin is also known as SYNPO. The spine apparatus (SA) is a specialized form of endoplasmic reticulum (ER) that is found in a subpopulation of dendritic spines in central neurons. The SA consists of a series of stacked discs that are thought to be connected to each other and to the dendritic system of ER-tubules. The actin binding protein synaptopodin (which has originally been described in podocytes of the kidney) is an essential component of the SA. Mice that lack the gene for synaptopodin do not form a spine apparatus. The SA is believed to play a critical role in learning and memory. In summary, an important function of the spine apparatus is the regulation of plasticity at individual synapses, a process known as metaplasticity. The International Radiation Hybrid Mapping Consortium mapped the SYNPO gene to chromosome 5.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Question

We are currently using anti-Synaptopodin/SYNPO antibody RP1069 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it possible that the antibody can work on horse tissues as well?

Verified Customer

Verified customer

Asked: 2019-11-12

Answer

The anti-Synaptopodin/SYNPO antibody (RP1069) has not been validated for cross reactivity specifically with horse tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in horse you can get your next antibody for free. Please contact me if I can help you with anything.

Question

Will anti-Synaptopodin/SYNPO antibody RP1069 work on primate WB with brain kidney?

Verified Customer

Verified customer

Asked: 2019-06-05

Answer

Our lab technicians have not tested anti-Synaptopodin/SYNPO antibody RP1069 on primate. You can run a BLAST between primate and the immunogen sequence of anti-Synaptopodin/SYNPO antibody RP1069 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in primate brain kidney in WB, you can get your next antibody for free.

Asked: 2018-04-18

Answer

The immunogen of RP1069 anti-Synaptopodin/SYNPO antibody is A synthetic peptide corresponding to a sequence at the C-terminus of human Synaptopodin (629-658aa EKPKVTPNPDLLDLVQTADEKRRQRDQGEV), different from the related mouse sequence by one amino acid, and identical to the related rat sequence. Could you tell me which isotype you are interested in so I can help see if the immunogen is part of this isotype?
